It'll take over 200 lb-ft torque to loosen so do it before you raise the car, wheel still on the ground, E-brake on, jack in place ready to lift. The main trick is to push out the old bearings and press in the new ones without damage.

Replace the entire axle, or rebuild/regrease/install a new boot? If you are just doing a boot, or rebuilding either the inner or outer CV joint, then just pop it out of the transmission, and disconnect the hub assembly from the car. Sounds like a bad wheel bearing to me. Usually, the noise is louder when turning one way when the weight of the car is shifted onto the bad part. Grinding, vibration, rubbing sounds are all indicative, as is dependence on speed. However, it could be the CV as well. That usually has a clicking/clunking when turning one way more than the other.
